The lab takes a multidisciplinary approach to the primary sensory neurons that initiate pain and itch, and to the mast cells and neutrophils that interact with them. Work spans receptor discovery, in vivo imaging, and translation into new therapeutics — including as a scientific founder of two biotechnology companies.
Discovered the Mrgpr family of GPCRs as itch receptors in sensory neurons — including MrgprA3 for chloroquine-induced itch and MrgprX4 for bile-acid and bilirubin–driven cholestatic itch — opening new avenues for anti-itch drugs.
Showed for the first time that dedicated itch-sensing neurons (MrgprA3+) exist separately from pain neurons, resolving a century-old debate, and mapped how the spinal cord codes itch versus pain intensity.
Identified MrgprB2/X2 as the mast-cell receptor behind pseudo-allergic drug reactions, neurogenic inflammation, and post-stroke brain inflammation, and MrgprA1/A2 as neutrophil receptors that guard against infection.
Discovered Pirt and Pirt2 (Tmem100) as regulators of the TRPV1, TRPA1, and TRPM8 channels that govern heat, cold, and chronic pain sensitivity — revealing new targets for analgesics.
Created Pirt-GCaMP reporter mice for in vivo imaging of sensory neuron activity, uncovering mechanisms of chronic pain such as coupled and sympathetically-driven cluster firing of sensory neurons.
Engineered CD200high human stem cell–derived nociceptors (hPSC-NNs) that, transplanted into osteoarthritic joints, sequester inflammatory ligands, promote bone and cartilage repair, and relieve pain — a disease-modifying cell therapy for chronic pain where conventional mono-targeted analgesics fail.
